Due to side effects, it's suggested that the starting dose be 25mg and as low as 12.5mg if side effects persist. As for what is the lowest effective dose, that depends on your physiology.

Supposedly, taking it in the morning should be effective all day. It is for me, but I have read reports from others here who seem to metabolize it more quickly. The "official" Eskapa word is that a single 50mg dose should still be effective at night even if you take it in the morning.
